20-07-07: Earthquake!

This afternoon my wife go shopping with another girl from Sichuan. It is surprising to see the SWAROVSKI is having 80% off. That never happens in China. She is going to buy a tiara for the wedding. In fact last Sunday I have seen it, and she tried it on as well. It looked really nice. Whatever, she bought a sheep named Shady (which she insists is a poodle) in the end.

I think the purification of Fd mut again and agree with Richard. The protein found in the binding buffer is too much and there may be more in the flow through. Nevertheless, I hope Christina will redo this as soon as possible and we could be sure of it.

This weekend will be busy, I may purify the Ed domain this Satuarday or Sunday, I have to... anyways.

My wife cooked some nice fish yesterday, brilliant.

18-07-07: Lab meeting day, disappointing football, arcade stick

Lab meeting is good, ehh I am not saying something like "the culture revolution is good, is good, is really good.". Since Richard is our supervisor, he is responsible to give adivce, isn't he? I was bothered by the low concentration resulted in Fd mutant protein purification and Christina has no idea on it either. Richard's opinion is that the mutation may changed the structure of the protein and the His-tag may be hidden.
I should describe the purification result: flowthrough has bands of the incorrect size, binding buffer has thin band of the correct size but since there is only 25mL of it the problem is small, washing buffer has no band at all, and elute buffer has thin band of the correct size. From this I suggest the protein concentration in the original solution is low or the protein was not eluted from the beads. The TCA result from Chirstina however indicates that there was enough protein in the original solution. So if this result is not wrong, and there is nothing wrong during the dialysis (not likely), the problem should be on the eluting process.
My beads truned yellow, I don't know whether this is related to this result or not. I may discuss this with people tomorrow.

I am planning to buy an arcade stick. Kofkofkof (or 3k) advised me to buy Real Arcade Pro V2.0 by Hori. He is such a honourable man so I guess I will buy one. The price is affordable, I may ask pearshaw to help me on this.
Talking about pearshaw, I got news about Diaquick. Diaquick is graduated, successfully, finally. He is clever, he should have been, so wish him a good future.

17-07-07: iPhone

Eh... What to say? I admit I have bias against Apple (you know what Apple, the company produced Apple computer). I don't like the Apple design for Mac, Mac book, iPod, etc. Those things just look way too foolish, I can not imagine how a man would like them. The Apple product nowadays is typical look nice, de facto malfunctional. So here I would like to give this link about iPhone.
It never worths it. I can understand the reason some girls would like it, but the price is too high.
In addition, I should say, why iPod is successful is because it constructed a legal mp3 downloading system. This is clever, but the iPod itself is not worth buying. Especially in China, whilst most people download pirate mp3s, there is no advantage for them to buy an iPod at a cost around 3,000 RMB.
Maybe the iPhone 2 will be much better, but not iPhone.

16-07-07: PhD programme, football shoe, old friends' blog

I left the westernblot membrane in PBT in 4oC cold room overnight yesterday. This morning I ... fluoresced (what is this stupid grammar here?!) it... with ECL, and it look nice, no worry.

Miyan never replies me, I am thinking of call him directly. However, Dr. Brown replied my mail today (I sent him the mail this morning), and we may have a talk in this week. What is funny is that he doesn't provide reference so I have no idea for this project at all - as well as the basic knowledge! Nevertheless, I will go.

I bought a pair of Umbro football shoes yesterday, only 15 pounds - cheaper than in China. It looks nice, I will try it out this Thursday. I will wear my glasses when playing this time definitely.

I took a look at some of my old friends' blog - so many guys are writing blog now! And those guys look just the same as before, maybe 5 years is too short for a person to change, maybe.

I will modify my report's introduction, the structure should be optimised.

Solid (or you can call his name Shen Zhouliang) is now on an evection, I sent him the download link for the CHD and clone mirror of SF 3.3. Hopefully he will be able to play that with me in the future.

15-07-07: weekend

I was in the lab yesterday till 6:30, largely because of my own fault...
Whatever, someone works much harder than me. Just as I said before, I am always a pupil, learning from everything and everyone. I will never be a master, be a instructor I guess. All I want to do is to learn, learn more.

There is one thing good to mention, I got the CHD file for SF 3.3 so I can play it on internet and fight against human opponents now. Just in case, I put the link here: sfiii3 chd, sfiii3 japan no cd rom. These storage will be expired soon.

Hope this afternoon the weather would be clear, my wife wants to shopping - I don't like rain shopping, neither does she.
